Augmentin Detailed Information
Augmentin is an oral antibacterial combination consisting of the
semisynthetic antibiotic amoxicillin and the beta-lactamase inhibitor,
clavulanate potassium (the potassium salt of clavulanic acid). Amoxicillin
is an analog of ampicillin, derived from the basic penicillin nucleus.
Clavulanic acid is produced by the fermentation of Streptomyces
clavuligerus. It is a beta-lactam structurally related to the penicillins
and possesses the ability to inactivate a wide variety of beta-lactamases
by blocking the active sites of these enzymes.
Augmentin is used to treat infections of lower respiratory tract,
otitis media, sinusitis, skin and skin structure infections, UTIs,
and community-acquired pneumonia caused by susceptible microorganisms.
The two components of Augmentin, amoxicillin and clavulanic acid
are fully dissociated in aqueous solution at physiological pH. Both
components are rapidly and well absorbed by the oral route of administration.
Absorption of Augmentin is optimised when taken at the start of
a meal.

Augmentin Advantages:
- May be taken with milk, juice, or food
- Broad spectrum of antibacterial activity against many gram-positive
and gram-negative micro-organisms
- The presence of clavulanic acid protects amoxicillin from degradation
by beta-lactamase enzymes and effectively extends the antibacterial
spectrum of amoxicillin to include many bacteria normally resistant
to amoxicillin and other penicillins and cephalosporins
- Good activity against the main pathogens associated with acute
otitis media (AOM) (penicillin-susceptible and -intermediate strains
of Streptococcus pneumoniae, and beta-lactamase producing strains
of Haemophilus influenzae and Moraxella catarrhalis) 4
Augmentin Disadvantages:
- High incidence of diarrhea 2
- Prolonged use may result in fungal or bacterial superinfection
- A high percentage of patients with infectious mononucleosis
have developed rash during therapy
- May cause false test results with Clinitest® urine glucose
monitoring
